Composition mixture -crotonolaktone and Zn-carnozine derivatives gel sanitization action at the experimental infected wound process

Abstract

The aim of the performed work was to present research results of antimicrobial and wound healing properties of a new medication gel, made on the basis of composition mixture -crotonolaktone and Zn-carnozine derivatives at the model of an infected dermatomic wound.

An experimental wound was designed according to standard methods of Ì. Abdulaiv with the help of the rounded stencil by general area of 120 mm2 (diameter – 12,5 mm).

All animals (white rats with weight of 180–220 g) were divided into three groups: control animals, for which treatment was not conducted; 1-st experimental group where on the wound surface 2% composition of mixture gel was applied; 2-nd experimental group where on the wound surface ointment “Oflacin-Darnytsa” was applied. Research was conducted on 3, 5, 7, 10, 14 and 21 day after forming of the wound. For determination of antimicrobial activity efficiency of the investigated mixture gel, material from the wound surface was taken by a sterile non-permanent tampon. In 24 hours to incubation counted up the amount of colonies which grew, and the result was expressed by the number of colony-forming unit on 1 ñm2 of area of the wound.

Findings testify that the offered medication owns bactericidal property on the wide spectrum of pathogenic gem flora for 2% concentrations of composition mixture. As a result, middle terms of stopping of selection of clean culture of staphylococcuss in the experimental group present 7,9±0,73 days, while in the group without treatment 12,7±0,48 days, and at treatment with “Oflocain-Darnitsa” ointment – 9,8±0,63 days.

Besides that, this composition mixture has another characteristic – stimulation of regeneration of dermatomic wound, especially in the initial periods of its cicatrization. It has been educed that its application promotes intensive liquidation of the sharp inflammatory phenomena and strengthening of necrolysis processes, as a result there is more rapid change of catastatic processes in the wound in anabolic, and that is reflected in complete wound healing in earlier terms.

Already in the first phase of wound process there was considerably more rapid liquidation of the sharp inflammatory phenomena, when massive exudate excretions cease (9,5±0,84 days in a control group a to 5,1±0,73 in experienced group).

Stopping of inflammatory process assists the speed-up passing of cicatrization process to the phase of necrolysis and cleaning from the necrotizing masses. If in the control group the indicated phenomena were observed only on a 11,7±0,67 days, then in the experimental group they are already present on 6,6±0,51 days.

Next phase is beginning of development of granulation reparative tissues naturally, as a result of more rapid previous, passes ahead in the experimental group control indexes and present according to 9,3±0,48 days to 13,7±0,67. However, it is necessary to pay attention to certain slowness of reparative processes in the experimental groups. If difference between beginning of previous and it phases in the control group, then it was presented on 2 days, while in the experimental it increased to 3 days. Similar dynamics was observed in next phases, where there remained difference between the beginning of every next phase in the control and the experimental groups.

It is also necessary to mark, that the composition mixture gel under study appeared effective at comparison both with the control group and with the group, where analogical treatment was conducted by traditional modern means – ointment “Oflocain-Darnitsa”.

In conclusion it is necessary to state that composition mixture of -crotonolaktone and Zn-carnozine derivatives owns expressive wound healing properties at its application in vivo and has a prospect for successful treatment of the infected wounds of soft tissues.
