Expression of mRNA of NLRP3-inflammatory cytokines IL-1β, IL-17α of small intestine in rats under chronic social stress

Abstract

Досліджували рівень експресії мРНК прозапальних цитокінів IL-1β, IL-17α та NLRP3– інфламасоми в кишково-асоційованій лімфоїдній тканині у щурів лінії Wistar за умов хронічного соціального стресу. Тотальну РНК виділяли за допомогою Trizol RNA Prep100 (Ізоген Lab., LTD, Росія), для проведення зворотної транскрипції й отримання кДНК використовували набір ОТ-1 “Синтол” (Росія). Для визначення рівня експресії мРНК генів IL-1β, IL-17α та NLRP3 проводили ЗТ-ПЛР в реальному часі на ампліфікаторі CFX96 ™ Real-Time PCR Detection Systems (Bio-Rad Laboratories, Inc., США). Відносний рівень експресії генів оцінювали за методом ΔΔCt, нормалізуючи за референс-геном GAPDH.

З’ясовано, що ХСС супроводжується транскрипційною індукцією генів прозапальних цитокінів IL-1β і IL-17α, а також NLRP3-інфламасоми в КАЛТ щурів, більш виразної у разі ХСС1. Проведене дослідження показало односпрямовану динаміку зростання транскрипційної активності генів NLRP3-інфламасоми (в 17 разів (p < 0,05) при ХСС1 і в 2,2 разу (p < 0,05) при ХСС2); IL-1β (у 6 разів (p < 0,05) при ХСС1 і в 2,8 разу (p < 0,05) при ХСС2); IL-17α (у 2,3 разу (p < 0,05) при ХСС1 і на 50 % (p < 0,05) при ХСС2) порівняно з контрольною групою. Проведені дослідження демонструють, що хронічний соціальний стрес суттєво підвищує рівень прозапальної сигналізації в кишківнику і може бути тригером розвитку запальних та автоімунних захворювань.
