Functional disorders of the central nervous system and their correction under conditions of separate and simultaneous exposure to lead and fluoride (experimental studies)

Abstract

Abstract. Lead and fluoride enter the human body orally, either separately or simultaneously, with food and water, and they are characterized by polytropic action, in particular disrupting the function of the CNS. Lead has a neurotoxic effect, causing encephalopathy and polyneuropathies, and decrease in intelligence. Prenatal exposure to fluoride increases the risk of developing neurobehavioral reactions, cognitive development indicators. Correction of the adverse effects of lead and fluoride on the functional indicators of the CNS by means of the sequential use of a complex of bioprotectors has not been studied sufficiently. Aim of the study: to investigate the long-term effect of lead, fluoride and their simultaneous action on the dynamics of subthreshold impulse summation and the startle reflex of white rats without the use of bioprotectors and following sequential addition of pectin, calcium and antioxidants to the standard diet of the animals. Materials and methods. Four series of studies were conducted on white rats, each based on the orthogonal planning scheme 22. Aqueous solutions of Pb(NO 3 ) 2 and NaF were administered separately and simultaneously into the stomach for 30 days: in series I without bioprotectors, in series II on the background of pectin, in series III – pectin and calcium, in series IV – a complex of pectin, calcium and antioxidants, vitamins C and E, β-carotene, selenium. On days 3, 15 and 30, the summation threshold index and the startle reflex, which characterizes the orienting and exploratory behavior of laboratory animals, were determined. Results. Separate and simultaneous exposure to lead and fluoride leads to a decrease in the ability of the CNS to sum subthreshold impulses and the orienting-exploratory behavior of laboratory animals due to the direct and indirect effect of CNS damage. Wave-like and multidirectional changes in indicators during the sequential addition of pectin, calcium and antioxidants to the diet characterize the phasic nature of inhibitory, excitatory and adaptive processes, quantitative changes characterize the strength of the action of lead, fluoride and their combination. Bioprotectors aimed at removing chemicals from the body and the mechanism of their action contribute to the recovery of functional disorders of the CNS. Conclusions. A complex of pectin with calcium and antioxidants can be considered the optimal means of correcting the summation of subthreshold impulses and orienting and exploratory behavior of laboratory animals during separate and simultaneous exposure to lead and fluoride.
